Biles, who has said she hopes to win five gold medals, took first place with a 15.966 average from her two vaults.
She has also won gold in the all-around and team competitions, and will now target wins on the beam and the floor.
Russia’s Maria Paseka (15.253) took the silver and Switzerland’s Giulia Steingruber (15.216) won bronze.
Uzbekistan’s Oksana Chusovitina, 41, who was looking to become the oldest gymnast to win an Olympic medal, finished seventh out of eight finalists in her seventh Olympic Games.
Chusovitina won a gold medal at her first Games in Barcelona in 1992 – five years before Biles was born.
